遺伝性前立腺疾患発症リスクに関わる単塩基遺伝子多型および責任遺伝子の同定

Research Abstract

2年間の研究期間の2年目である平成17年度では、単塩基遺伝子多型(SNP)のスクリーニングを引き続き行った。家族性前立腺癌の責任遺伝子として同定されたMSR1遺伝子は非家族性前立腺癌の発症リスクとなるが、この遺伝子について日本人の家族性および非家族性前立腺癌および非前立腺癌患者において解析を継続した。日本人に特異的なSNPは同定されなかったが欧米で前立腺癌感受性とされたSNPの頻度が日本人では異なっており、前立腺癌発症の人種差を説明する一つの要因であることが示された。また、やはり家族性前立腺癌の責任遺伝子の一つであり非家族性前立腺癌発症リスクとなるRNASEL遺伝子についても同様の検討を行ったがこちらでも日本人に特異的なSNPは同定できなかったものの、欧米で前立腺癌感受性遺伝子とされたSNPの頻度が日本人では異なっており、前立腺癌罹患率の人種差を説明する一つの要因であることが示された。これらの成果については現在投稿準備中である。
米国ジョンズホプキンス大学病院泌尿器科のWilliam B.Isaacs教授の研究室との共同研究で行ったKLF6遺伝子についてその解析結果を報告した(Narla G and Komiya A et al., Cancer Research : 2005)が、日本人についても解析を行ったものの、この遺伝子については有意な結果は得られていない。
以上の結果から欧米で家族性前立腺前立腺癌より同定され、前立腺癌感受性遺伝子とされている遺伝子群のSNPの頻度は日本人の家族性および非家族性前立腺癌では異なっており、疾患発症リスクをあげるとされるSNPは日本人の間では有意な結果は得られていない。これらから、日本人の前立腺癌発症のメカニズムは欧米とは異なっているということが推察されるがこのメカニズムの解明は今後の課題である。
