カドヘリンノックアウトマウス痛覚回路のin vivoパッチクランプ解析

Research Abstract

In vivoパッチクランプ法や脊髄スライスパッチクランプ法および免疫組織化学的手法を用い、脊髄後角第II層、膠様質を介した痛覚伝達路に特異的に発現する接着分子、カドヘリン8が痛みのシナプス伝達に対して如何なる機能的役割を果たすかを解析した。正常動物の膠様質細胞からin vivoパッチクランプ記録を行い、皮膚へ機械的痛み刺激や触刺激を加えると、EPSCの発生頻度や振幅が著明に増大した。一方、熱刺激に応答を示す細胞は見られなかった。カドヘリン8ノックアウト動物でも同様の解析を行い、EPSC(興奮性シナプス後電流)の発生頻度や振幅を正常動物から得られた結果と比較した。その結果、機械的痛み刺激によるEPSCの発生頻度がノックアウト動物では減少した。触刺激や熱刺激の応答には差は得られなかった。次に、記録細胞を染色すると、約15%の膠様質細胞がカドヘリン8を発現し、これらの細胞は主に皮膚へ機械的痛み刺激によってEPSCの発生頻度と振幅を著明に増大した。また、抑制性介在ニューロン(GABAなどの伝達物質を含有する)であるislet細胞にはカドヘリン8の発現は観察されず、興奮性細胞(グルタミン酸を伝達物質とする)であるstalk細胞などにその発現がみられた。電子顕微鏡による観察から、膠様質細胞とC線維との特徴的なシナプス結合様式の一つであるglomeruliにおけるシナプス外側部にカドヘリン8が観察された。前年度の研究から、ノックアウト動物における機械的痛み刺激に対する逃避行動閾値の減少、膠様質細胞へのC線維を介したシナプス入力の減弱が見出された。
以上本研究により、カドヘリン8は、機械的痛みを伝達するC線維とstalk細胞などの興奮性の膠様質細胞とのシナプス形成に必須であり、それを欠損すると、そのシナプス入力の減弱に伴う機械的痛みに対する行動の異常が惹起されることが明らかになった。
