Research Abstract

亜硝酸還元酵素(NIR)は、脱窒過程の一部である亜硝酸イオンを一酸化窒素に還元する反応を触媒する酵素である。NIRのうち、通常の銅型NIRはサブユニット(単量体)あたり電子受容部位として働くタイプ1銅と、亜硝酸還元部位として働くタイプ2銅の二つの銅結合部位を持ち、3量体構造をとることが知られている。これに対してHyphomicrobium denitrificans A3151株由来の銅型NIR(HdNIR)は単量体あたり2つのタイプ1銅(type 1 Cu_N,type 1 Cu_C)と1つのタイプ2銅を持ち、6量体構造をとる。
HdNIRは、そのアミノ酸配列のN末端側にブルー銅タンパク質のプラストシアニンと相同性のあるドメイン(N-terminalドメイン)とC末端側に既知のNIRと相同性のあるドメイン(C-terminalドメイン)からなり、Nドメインにはtype 1 Cu_N、Cドメインにはtype 1 Cu_Cとtype 2 Cuが含まれている。これまでにNativeおよびC260Aの結晶化を行い、2.4Å分解能までのX線回折強度データを収集した(Acta Cryst.,D60,2383-2386(2004))。銅イオンを利用したMAD法による構造解析を試みたが、最終的には、分子置換法に成功し、2.4Å分解能でR値18%の精密構造を得た。
構造解析の結果、HdNIRのN末端ドメインのtype 1銅の周囲に存在する、Gly74,Gly38,Ala75,Met40,Ile35などの疎水パッチ同士が結合し、3量体構造どうしがface-to-faceで向き合うような構造を有していた。NIRドメインのtype 1とtype 2の銅イオンの位置は、12.7Å離れ、N末端ドメインのtype 1と、NIRドメインのtype 1とは50.4Å離れていたが、隣のNIRドメインのtype 1との距離が24.7Åと近く、異なる分子への電子伝達経路が考えられた。
ストップドフローやパルスラジオリシスなどの生化学実験の結果から、HdNIRの生理的電子供与体であるシトクロムc_<550>は、2系統の電子伝達経路でHdNIRに電子を供与し、N末端ドメインのtype 1銅に電子を渡す速度よりも、NIRドメインのtype 1銅へ電子を渡す速度が非常に速いことが判明した。これら2つの電子伝達経路を構造から議論した。
