Research Abstract

(1)葉緑体ゲノムを用いたマルチカラーFISHによる性染色体構造解析
昨年度の性染色体特異的BACクローンの解析により、特異的BACクローンBAC43は葉緑体ゲノムの一部をコードしていることが判明した。ビオチン標識した葉緑体ゲノムをFITCで、DIG標識したX43.1をローダミンで、染色体をDAPI染色で検出したマルチカラーFISHを行った。葉緑体ゲノムはX染色体よりもY染色体に特異的なシグナルが検出された。このことから、ヒロハノマンテマのY染色体には葉緑体ゲノムが特異的に蓄積していることが判明した。この研究成果を国際学術雑誌Geneticaに投稿し下記のように受理された。

(2)雄性生殖器官特異的遺伝子SlMF1の性染色体局在解析
雄性生殖器官特異的遺伝子SlMF1の発現解析をin situ hybridization で行い、雄蕊に特異的に発現していることを明らかにした。そこで、ヒロハノマンテマのアメリカの系統および日本の系統を用いてゲノミックサザン解析を行い、SlMF1のコピーの内、少なくとも1コピーがY染色体に局在することが明らかになった。さらにX染色体を用いたフローソーティング解析により、SlMF1はX染色体にも存在することが明らかになった。以上の研究成果を国際学術雑誌Genes & Genetic Systems に投稿し下記のように論文を出版した。
