Research Abstract

選択的リグニン分解菌Ceriporiopsis subvermisporaは,木材腐朽初期にリノール酸などの不飽和脂肪酸を大量に産生する.この不飽和脂肪酸は,選択的リグニン分解機構に重要な脂質過酸化反応に関与するものと予想されることから,今回は,C.subvermisporaより脂肪酸の不飽和化に関与する酵素(fatty acid desaturase)の遺伝子クローニングを試みた.
既知のfatty acid desaturaseのアミノ酸配列の中で高度に保存された領域から縮重プライマーを設計し,ゲノムDNAを鋳型としてPCR,また,total RNAを用いて5'-及び3'-RACEを行った.その結果,delta9-fatty acid desaturase(Cs-Fad1と命名)とdelta12-fatty acid desaturase(Cs-Fad2)の完全長のcDNAを取得した.これら遺伝子がコードするアミノ酸配列には,既知のfatty acid desaturaseに特徴的なhistidine-rich motif(His-box)がそれぞれ存在した.また,SOSUI解析の結果,(Cs-Fad1及びCs-Fad2は膜結合型酵素であることが強く示唆された.さらに,Cs-Fad1については,C末端側にcytochrome b_5 heme-binding domainが存在した.今回取得したCs-Fad1及びCs-Fad2を昨年度取得した白色腐朽菌Phanerochaete chrysosporiumのPc-Fad1及びPc-Fad2と比較したところ,アミノ酸レベルでそれぞれ約75%程度の相同性があったが,膜貫通ドメインの個数などに違いが見られ構造が異なっていた.
