Research Abstract

MAPK,特にERKの活性化は蛋白質のリン酸化といった早期における神経系の可塑的変化だけではなく,遺伝子発現レベルでの制御といったlong termにおける可塑的変化にも関与している.一方,近年,次々と発見されている温度感受性イオンチャネルはTRPスーパーファミリーの一部として整理されている(熱刺激受容体であるTRPV1,TRPV2,温刺激受容体であるTRPV3,TRPV4,冷刺激受容体であるTRPM8,TRPA1)。我々は様々な慢性痛モデルを用いて,一次知覚ニューロンにおけるTRPM8,TRPA1遺伝子発現の変化と慢性疼痛との関連を検討した.これらTRPチャネル温度受容体は一次知覚ニューロンの温度受容のメガニズムを解明する上で極めて重要な分子であり,これまでは熱刺激受容体であるTRPV1,TRPV2を中心に,主に熱性痛覚過敏との関連について研究が進められてきた.しかしながら,一次知覚ニューロンにおける冷刺激受容体の発現変化と,慢性疼痛の病態との関連は現在までのところ全く不明であった.まずMAPKの一つであるp38 MAPKの活性化が,TRPA1を遺伝子発現レベルで調節し,末梢炎症に起因する慢性疼痛の病態に関与していることを明らかにした.またこの変化がNGFのpositive regulationによって生じていることが分かった.一方,部分的軸索損傷モデルである神経因性疼痛モデルの傷害DRGニューロンにおいては,ERK, p38,及びJNKすべての活性化がみられるが,非傷害DRGニューロンにおいてはp38のみ活性化されることが分かっている.この非傷害DRGニューロンにおけるp38の活性化が,末梢炎症モデルと同様に,TRPA1の発現上昇を介して冷刺激に対する痛覚過敏に関与していることを見いだした.このようにDRGニューロンにおけるTRPチャネルは様々な病的状況下において,p38 MAPKをはじめとする細胞内シグナル伝達によってその発現を調節されており,この発現変化が慢性痛の発生に関与していることが明らかになった.これらの成果はJ.Clin.Invest.やPain等、数多くの国際誌に発表した.
