Research Abstract

糖尿病状態で促進的に形成される後期糖化反応生成物(advanced glycation endproducts、AGE)がAGE特異受容体(receptor for AGE、RAGE)を介して糖尿病合併症の発症・進展に関わることが明らかになってきた。RAGEは一回膜貫通型受容体で、その細胞外領域に2本のN-結合型糖鎖構造をもち、これらの糖鎖付加部位に挟まれたごく狭い領域がAGEリガンド結合部位である。本研究の目的は、RAGEの糖鎖修飾がリガンド結合に与える影響を明らかにすることである。
(1)培養血管内皮細胞株ECV304細胞に、野生型、それぞれ片方の糖鎖付加部位(-NXS/T-)のNをAにした変異導入型、両変異導入型の計4種類の組み換えRAGE蛋白を強制発現した。N末端側から2番目のN型糖鎖結合部位への変異導入によって、AGE-BSAに対するVEGF mRNAの発現増強が生じた。
(2)培養血管内皮細胞で発現するRAGEの糖鎖修飾には、高血糖、AGE刺激、インスリン負荷のいずれにおいても、あるいは3者同時負荷においても影響が生じなかった。しかし、50mg/dl以下の低血糖暴露で糖鎖修飾がほぼ完全に阻害されることが明らかになった。
(3)糖尿病あるいは非糖尿病正常マウスの脳、肺、腎、小腸からタンパクを抽出し、RAGE糖鎖修飾の有無をウエスタンブロットで推定した。その際に、N型糖鎖を酵素的に外すglycopeptidase-Fを使用した。腎臓においては糖鎖修飾のないRAGEも発現していた。
RAGEのN-結合型糖鎖付加修飾がないと、AGEリガンドのRAGEへの結合能が亢進することが明らかとなった。低血糖暴露がRAGEのN-結合型糖鎖付加修飾を阻害する一因子であった。RAGEのリガンド結合部位の糖鎖付加修飾の多様性が糖尿病血管症の発症・進展に関わることがはじめて分子レベルで証明された。
