Research Abstract

本研究は、癌が血行動態以外に臓器特異的な転移をすることに着目し、癌が転移する前に原発巣より影響を及ぼすと思われる宿主側の臓器の遺伝子、蛋白レベルでの発現の変化をスクリーニングし、標的分子を同定する目的である。担癌マウスをもちいた転移前の肺の遺伝子レベルでのスクリーニングの為、癌細胞(Lewis lung carcinoma及びB16 melanoma細胞)を皮下に移植し腫瘍を形成させる。この担癌マウスの腫瘍が増大して肺に転移を起こす前の肺を採取し、癌を有していない正常のマウスの肺と比ぺ、遺伝子レベルで発現に差のあるものをmouseのチップを用いたマイクロアレイでスクリーニングした。候補遺伝子のうち、S100ファミリーのS100A8とS100A9は、転移前にmRNAレベル、蛋白レベルともに肺の血管内皮細胞、マクロファージに強い発現誘導が認められた。さらに担癌マウスの肺におけるこの2分子の上昇が、転移と関与するかについて検討する為、中和抗体の作製を試みた。S100A8とS100A9は腫瘍細胞に対して遊走活性を引き起こすことを明らかとし、中和抗体の一つがin vitroでこの機能を抑制することが可能であることが分かった。この抗体をin vivoに投与したところ、転移の抑制が認められた。このことより、転移前の肺における走化因子の発現を抑制することにより、早い段階での肺転移の予防的治療の可能性が示唆された。
