Research Abstract

Gab2.WT Tgマウス、Gab2.deltaPro (Grb2/Gads SH3ドメインに結合するproline rich領域を欠損させたもの)TgマウスをそれぞれMHC class II拘束性のOH-II TCR Tgマウス、MHC class I拘束性のH-Y TCR Tgマウスと交配させ、胸腺におけるT細胞選択に及ぼすGab2の影響を調べた。Gab2. WT Tgマウスでは、OT-II TCRにおけるpositive selectionが顕著に減少したが、Gab2.dPro Tgでは正常であった。H-Y TCR♂で観察されるnegative selectionも、Gab2.WT Tgマウスで抑制されたがGab2.dPro Tgでは正常であった。この結果より、Gab2はpositive selection, negative selectionの負の制御因子として働き、その機能にはproline rich領域が必須であることが判明した。H-Y TCRのシステムは全身性抗原によるnegative selectionと定義されるが、近年末梢抗原(組織特異的抗原)によるnegative selectionは胸腺上皮細胞において全く異なるメカニズムで起こっていると考えられている。そこで、ラットインスリンプロモーターでOVAを発現するTgマウス(RIP-mOVA Tg)とOT-I,OT-IIとのdouble Tgによるnegative selection modelを作製し、組織特異的抗原に対するcentral tolerance獲得過程におけるGab2の役割を検討中である。
