Research Abstract

ヒト骨髄由来間葉系幹細胞(hMSC)は間葉系細胞・組織へ分化するだけではなく、皮膚の創傷治癒においても重要な働きを示す(Nakagawa H, Akita S, Akino K, et al. Br J Dermatol, 2005)。一方、皮膚と同様に同じ外胚葉系の神経系への間葉系幹細胞の分化が示されており、hMSCの神経系への分化、関与を神経ホモログを持つShc関連遺伝子・タンパク発現、Stathmin関連遺伝子・タンパク発現を確認し、Shcのノックアウトマウスを用いて、創傷治癒へのin vivoでの関与、ノックアウトマウス由来骨髄間葉系幹細胞(間質細胞)の増殖性、分化能につき検討した。hMSCの免疫細胞染色では、ShcA、ShcC、SCG10、Stathmin等のタンパク発現を認めたものの、ShcB発現は認められなかった。Semi-quantitative RT-PCR法による遺伝子発現検討では、ShcA、ShcC、SCG10、SCLIPの発現同定したもののShcB、RB3は発現認めなかった。Shcタンパクは細胞内アダプタータンパクとしてMAPK/Erk系を介して増殖・分化に関与すると思われるので、ShcCノックアウトマウス由来骨髄間葉系幹細胞(間質細胞)の増殖性とノックアウトマウスin vivoでの創傷治癒を検討したところ、細胞増殖性は4日間観察において対照は5.5倍に増殖したものの、ノックアウト由来細胞は1.5倍に留まった。リン酸化MAPK/Erk発現では44及び42Kdaにほぼ1/10の発現量であった。10×10cm^2の全層欠損創において、人工真皮(ペルナック【○!R】)被覆観察6日で、対照littermateは完全創傷治癒したものの、ノックアウトマウス創は全く治癒しなかった。正常littermateとShcCノックアウトマウスの皮膚構造を比較したところ、ノックアウトマウスでは表皮がほぼ単層であり毛包、脂腺が分離し、共に未熟な構造を取っていた。また、正常littermateでのShcC、ShcA発現は真皮発現しており、表皮には認められなかった。
