Outline of Annual Research Achievements

本研究では、合成ポリマーの側鎖に種々のアミノ酸を結合させ、トランスポーターとの相互作用を介してがん細胞と特異的に相互作用する高分子リガンド分子の開発を目指して研究を実施してきた。分子量の異なるポリアミノ酸の側鎖に、トランスポーターに認識される形で種々のアミノ酸を結合し、がん細胞および正常細胞との相互作用を評価するスクリーニング試験を行った結果、がん細胞において過剰発現していることが知られているトランスポーターXに特異的に結合するアミノ酸Yを結合したがん細胞特異的高分子リガンドを得ることができた。高分子リガンドのがん治療への応用に関しては、ポリアミノ酸の末端に光増感剤IRDye DX700（水溶性フタロシアニン誘導体）を結合し、光線力学治療（Photodynamic Therapy, PDT）への応用を検討した。DX700結合高分子リガンドを培養細胞と一定時間培養した後に、培地交換を行い、400-700nmのバンドパスフィルターを用いて光照射を行った結果、DX700単独と比較して顕著な光選択的細胞毒性を示すことが確認された。近年、光増感剤を結合したがん特異的抗体による光免疫治療が臨床試験で優れた治療効果を示し、大きな注目を集めているが、抗体の場合には特異的がん抗原を有するがん細胞にしか用いることができないことが課題である。これに対して、本研究では、DX700結合高分子リガンドの開発を行い、その有用性を明らかにした。本システムでは、がん細胞の生存と増殖に必要不可欠なアミノ酸を取り込むとトランスポーターを標的とすることで、抗体では困難なあらゆる固形がんの標的化が可能であり、イメージングや光線力学治療への応用が期待できる。
