Outline of Annual Research Achievements

本年度では、前年度で合成法を確立したモジュール の一つである、光学活性1,2-O,N型化合物を出発原料として、スフィンゴ新合成を行った。その結果、 Grubbsオレフィンメタセシスによるアリルアルコール合成、2度目の脱水型アリル化、再度のオレフィンメタセシス、脱保護による合成経路を確立した。モジュール合成を含めても5段階であり、現在の最短段階合成法である。脱水型分子内(不斉)アリル化、オレフィンメタセシスという同じ反応系を繰り返すだけの単純な戦略である。これをさらに繰り返せばより複雑な多官能性化合物への展開も期待される。本手法の有効性を示すことができた。
これに加えて、従来検証してきたアリルアルコールを用いる「脱水型」不斉アリル化反応から、単純なオレフィンを用いる「脱水素型」アリル化反応に焦点を移し、その反応系の検証を進めた。結果、独自に開発した光学活性ビスアミジン型配位子をもつ銅錯体にその有効性を見いだすことに成功した。酸化剤として取り扱い容易なジ-tert-ブチルペルオキシドを用いて、b-ケトエステル類をシクロアルケンと反応させると、アリル位のC-Hを切断しながら活性化メチレンとの付加反応が進行し、対応するアリル化化合物を与える。ジアステレオマー比は最大で99:1に及ぶ。未だ選択性向上に余地を残すが、第四級不斉炭素の構築も可能である。エナンチオ選択性を初めて示すことができたことは、今後の方向性を示すものとしてその展開が強く期待される。
