Outline of Annual Research Achievements

電気抵抗に基づいた多重チャンネルによるウイルスの検出のために、多機能性と多様性から注目を集めているグラフェンを用いた。グラフェンに金(Au)と酸化鉄のナノ粒子(NP)を修飾したAu-グラフェンを調製し、多機能プラスモングラフェン(dual-functional plasmomagnetic graphene、PMGRP)とした。このPMGRPは、磁性とプラズモン基質の両方の役割を果たし、本研究のマグネトフルオロイムノセンシング(MFIS)プラットフォームとした。プラズモンによる蛍光調節を検証するために、PMGRPと量子ドット(QD)に抗ヘマグルチニン抗体(HA Ab)を修飾した。インフルエンザウイルスの存在下では、PMGRPとQDの間のサンドイッチ構造(PMGRP-ウイルス-QD)を形成し、PMGRP上のAuとQD間距離が縮まり蛍光が増強した。蛍光強度はウイルス濃度に依存し、インフルエンザウイルスA(H1N1)は7.27 fg/mlの検出限界を示した。生物学的モデルとしてヒト血清とウイルスに混入したサンプルの場合には、磁力によって不純物から分離され、感度の低下がなかった。この検出法は、不純物を多く含んだ複雑な生体サンプル中のウイルスの検出には非常に有効であると考えられる。この原理によって、複数の抗体をPMGRPとQDに修飾することで、異なるウイルスの検出も可能であった。
さらに電気伝導性が良いカーボンナノチューブ(CNT)を用いることによって、電気化学的ウイルスの検出を検証した。Au-CNT複合ナノ材料に抗体を修飾し、静電力で白金電極に貼り付け、ウイルスの検出を行なった。標的ウイルスの存在下では複合ナノ材料の導電率変化が変化し、インフルエンザウイルスおよびノロウイルスについての検出限界は、それぞれ約8.4pMおよび8.8pMであった。したがって、この複合ナノ材料をベースした検出法は、DNAやウイルスの検出に優れた検出能力を示し、高感度で選択的な検出法として広く適用することができる。
