Outline of Annual Research Achievements

H29年度は、単一細胞の大変形に関する粘弾性の直接測定に挑戦した。その結果、これまで力の次元の無い相対値としてしか求まらなかった細胞の粘弾性を表す各係数を次元付きの値として定量化することができた。マイクロ流体デバイス内における直径8マイクロメートルの赤血球細胞に関する高スループット粘弾性測定は、力の測定機構、測定精度の両面でチャレンジングであったが、前年度に確立したマイクロ流体デバイス内細胞マニピュレーションを活用した幾つかの手法でこの課題に当たった。
試行錯誤の中で特に細胞の弾性係数測定手法として意義深い結果を得られたのが、マニピュレーション中に流体から受ける力を推定しヤング率に変換する手法と、形状ゆらぎ解析(flicker spectroscopy)を行い表面張力や曲げ弾性係数を得る手法である。マニピュレーションシステムと共存する形で上記の二種の測定手法に最適なイメージング法や実験条件を構築することで、赤血球細胞について、マイクロ流路内で4つの力学パラメーター(2つの弾性係数と2つの粘性係数)を単位付きで推定することに成功した。そこで得られた値は、いずれも過去に報告されている異なる手法と比較して適切なものであり、前年度に明らかにした変形能の変形時間依存性やATP濃度依存性などと合わせて細胞の生命ならではの力学特性を適切かつ高速に定量化することに成功しているといえる。これらの成果は国際会議のプロシーディングス論文として投稿中であり、今後実験・解析ともにさらに内容を深め、学術誌への論文投稿を行う予定である。
