Outline of Annual Research Achievements

本研究では,ジベレリン(GA)を介したエチレンによる果実形成の分子制御機構を組織レベルで解明することを目的とする.本年度は以下の研究および発表を行った.
1)未受粉でも子房が老化せずに種無し果実を形成するトマト単為結果性変異体3系統について,GAの関与を含む特徴付けおよび論文発表を行った.
2)前年度までに野生型,GAおよびエチレン変異体を利用し,レーザーマイクロダイセクションを利用したRNA-seqおよびWGCNAを行い,着果時のトマト子房における組織依存的転写ネットワークを明らかにした.これにより同定された,組織依存的発現パターンを示す4遺伝子の機能解析を目的とし,CRISPR/Cas9法によるトマトゲノムの編集を行った.その結果,GAやエチレンに関連する4遺伝子それぞれについて機能を完全に欠損していると考えられる植物体を得た.うちエチレンに関連する標的 1遺伝子の改変体について,着果に着目した解析(下記項目3,4,5)を行った.
3)標的遺伝子改変体の未受粉子房において2種類の老化マーカー遺伝子の転写発現解析を行った結果,いずれも野生型に比べ大きく低下していたことから,標的遺伝子改変体では未受粉の子房で生じる老化が抑制されていることが示唆された.
4)上記1)を含むこれまでの研究から,子房の老化抑制は,単為結果性の付与をはじめ,果実形成を促進する可能性が期待された.そこで,標的遺伝子改変体において開花前に除雄した花を観察した結果,単為結果性を示すことが明らかとなった.
5)未受粉子房の老化は,花が受粉および着果可能な期間を制限している可能性があると考えた.そこで,標的遺伝子の機能欠損が着果可能な期間を延長するか調査した.その結果,野生型と標的遺伝子改変体はいずれも開花4日後から6日後にかけて受粉による着果能力を大きく失い,受粉による着果可能な期間の延長は見られなかった.
