Outline of Annual Research Achievements

本年度まず申請者は、Flavodiironタンパク質 (FLV) およびFLVに関連した分子メカニズムの反応機構の解明に取り組んだ。まず一年目に大腸菌を用いて作製したFLVリコンビナントタンパク質を用いて、FLVの基質同定を目指した。実験に際しては共発現系を用いる事でFLV1およびFLV3のヘテロダイマーを精製する事に成功し、同時にFLVがホモダイマーではなくヘテロダイマーとして機能する事を支持する結果を得るに至った。当実験によって最終的には、FLVが従来考えられていたNADPHを基質とした酸素の4電子還元を触媒しない事が明らかとなった (Shimakawa et al. 準備中)。さらに申請者は、シアノバクテリアSynechocystis sp. PCC 6803においてFLV2/4と同オペロン上に存在するCO2応答性タンパク質であるColAに着目し、当該タンパク質を独立して保有するSynechococcus sp. PCC 7002を用いる事で、その生理的意義に新たな知見を見出した (Shimakawa et al. 2017 Marine Drugs 15, 390)。
また申請者は、FLVとは異なる光エネルギー散逸機構、つまり活性酸素傷害防御メカニズムとして、シアノバクテリアにおいて呼吸末端酵素が光化学系Iの光傷害緩和に貢献する事を初めて見出した (Shimakawa et al. 2018 Photosynthesis Research in press doi: 10.1007/s11120-018-0495-y) 。
平成29年度を通して申請者は、特別研究員の研究課題に係る3報の筆頭著者論文と1報の共同著者論文を研究業績として上げ、さらに国際分子珪藻学会および日本植物生理学会国際シンポジウムにおいて口頭発表を行った。
