糖代謝改善を目的とした新たな運動処方の開発 -IGF1に着目して-

Outline of Annual Research Achievements

本研究は,血糖コントロールに対するレジスタンス運動効果とその作用メカニズムの一旦を,IGF-1を含む特定の糖代謝制御因子に着目し,明らかにすることを目的としている.前年度までは,単回レジスタンス運動による骨格筋でのIGF-1発現増加が,糖尿病モデル動物においても同程度生じることを明らかにした.また,単回レジスタンス運動後の骨格筋への糖取り込み亢進に,IGF-1発現増加が貢献しているかを解明する遺伝子工学的手法を立ち上げた.今年度は,前年度立ち上げた実験系を用いて,予備的実験を行なった.この結果,本手法が安静時の骨格筋におけるIGF-1 mRNA発現の低下を引き起こし,単回レジスタンス運動によるIGF-1発現の増加を消失させ得ることが示唆された.さらに,レジスタンス運動後のIGF-1発現増加が,下流のシグナルに貢献している可能性が確認された.これらの結果から,レジスタンス運動による糖代謝の亢進には,骨格筋のIGF-1発現増加が重要である可能性が示唆された.次に,IGF-1刺激による糖取り込みを媒介すると考えられる因子(APPL1)に,慢性レジスタンス運動が与える影響についても検討した.その結果,慢性的なレジスタンス運動の実施が,APPL1の発現を増加させることが明らかとなった.さらに,この応答は健常時及び2型糖尿病罹患時の両条件で確認されることが明らかとなった.以上の結果より,単回及び慢性レジスタンス運動が血糖コントロールを亢進するメカニズムに上記因子が関わっている可能性が示唆された.
