Outline of Annual Research Achievements

本研究の目的は,脳内病理に対する治療薬の触媒的不斉全合成法の確立である。既に高い薬理活性が確認されているにも関わらず合成上の問題から研究が進んでいない化合物に着目した。例えばDCG-IV,Dysibetaine CPa,LY2140023,Ginkgolide B 等の構造的特徴である電子求引基が置換したシクロプロパン骨格やγラクトン縮環型骨格,tert-ブチル基を有する縮環型骨格に着目し,その高立体選択的な不斉合成法を確立し,目的の生理活性物質の簡易な新規合成法の確立を行う。また,量子化学計算を用いた反応機構解析を行う。
今年度は,DCG-IV,Dysibetaine CPaの大量合成法確立のため,反応条件の検討を行った。これまでの検討ではジクロロメタンを溶媒として用いていたが,今回,新たに開発した水溶性Ru触媒を用いてジエチルエーテル/水での二層系での反応を行った。その結果,36%収率,96%eeで目的のシクロプロパン化合物を得ることに成功した。2相系での反応では,分液操作のみで簡単に単離生成することができるというメリットがあり,大量合成において有用性が高いと考えられる。
またカルベン移動反応の高度な応用として,tert-ブチル基への位置および立体選択的カルベン挿入反応の開発を目的に基礎反応開発を行った。Ru触媒存在下で,原料のジアゾアセトエミド類は速やかに反応し,最高99%収率,91%eeで目的化合物が得られた。これは,tert-butyl基の1級C-H結合の高立体選択的な官能基化反応として初めての報告例である。また,Ru触媒の反応機構について量子化学計算を用いて解析を行い,不斉誘起機構を解明した。
以上の研究成果は,Journal of Organic Chemistry等に5報の論文として掲載された。
