Outline of Annual Research Achievements

本研究の目的は、誘導性制御性T細胞(iTreg)の免疫抑制に関わるエピジェネティック制御の分子機構を解明し,新たな免疫疾患治療法を開拓することである.
iTregはドナーからの細胞源採取が容易であり,かつアロ抗原特異的な誘導も可能であるため実際に臨床応用が強く期待されてきたが,Tregのマスター遺伝子であるFoxp3の不安定性が以前より課題とされており、実際の有効性や副作用についてまだ不明の点が多い.
我々はレシピエント型の抗原提示細胞とドナー型のナイーブT細胞の共培養からiTregを作成する際にDNA脱メチル化酵素であるTETの活性化物質であるビタミンCを用いたエピジェネッティック改変にてiTregの機能改善を目指し,その臨床効果と分子生物学的改変の意義に関する詳細について,急性移植片対宿主病(GVHD)モデルマウスやDNA脱メチル化解析を用いて明らかにした.Foxp3の生体内での高い安定性、炎症性サイトカインの産生抑制、Treg関連遺伝子の特異的DNA脱メチル化、致死的GVHDの臨床的な症状および生存曲線の改善などを確認できた。
またマウスのみならずヒトの細胞でもFOXP3highの安定した分画をVitCにより誘導することが出来ることが明らかになり、部分的ではあるがDNA脱メチル化も確認したことにより、本研究の臨床応用の可能性も広がったと考えている。
これらをInternational Immunology誌にまとめて発表し、本研究のpublicationとした。
