Outline of Annual Research Achievements

申請者は三年目に、ミトコンドリアの品質管理を人為的に制御する方法を模索した。卵子内のミトコンドリアは分解と合成のバランスの基にその量が制御されており、このミトコンドリア品質管理機構の存在は卵子の質を維持する上で重要である。申請者は卵子の質の向上を目的とし、卵子培養中の短期間の高温処理によりミトコンドリア品質管理が促進されると仮説を立て、検討した。
通常培養環境より3℃高い41.5℃にて1時間処理すると、熱ショックタンパク質であるHeat Shock Protein 72 (HSP72)の卵子内における発現が増加した。さらに処理後23時間が経過すると、卵子内においてミトコンドリア品質管理を制御しているSIRT1の発現が上昇していた。このSIRT1発現の上昇は、非高温処理のHSP72活性化剤処理卵子でも同様に観察されたため、高温処理がHSP72を介してSIRT1発現を制御することが示唆された。
次にミトコンドリア数を測定したところ、高温処理区と非処理区の間で差は見られなかった。しかし、卵子培養培地にミトコンドリア分解を行うプロテアソームの阻害剤(MG132)を添加したところ、非高温処理卵子と比較して高温処理卵子のミトコンドリア数が有意に増加した。このことから、高温処理卵子ではミトコンドリアの分解と合成の双方が活性化し、ミトコンドリアが作り変えられていることが示唆された。そこで高温処理卵子のミトコンドリア活性を調査したところ、ミトコンドリア膜電位活性とATP含量が有意に増加していた。このことから、短期間の高温処理は卵子内のミトコンドリア品質管理を促進させ、ミトコンドリアの質を総体的に向上させていることが考えられた。また、高温処理卵子では胚発生能力の向上が認められたため、この方法が卵子の質向上に有効であることが示された。
