Outline of Annual Research Achievements

体内の過剰な脂肪蓄積は動脈硬化度を増大させる大きな原因の1つであるが、有酸素性トレーニングによる体脂肪の低下に伴い動脈硬化度は低下することが報告されている。最近、脂肪組織から分泌される新規アディポカインとして複数のC1q/tumor necrosis factor-related proteins(CTRPs)が同定され、運動による動脈硬化度低下の新たなメカニズムにCTRPsが関与する可能性が考えられる。昨年度の研究成果として、中高齢者における運動による動脈硬化度低下には内臓脂肪だけでなく局所的な脂肪蓄積の変化が関与すること、その機序に体脂肪の減少に伴う血中adiponectin、CTRP3、CTRP5濃度の増加が関与する可能性が明らかとなった。しかしながら、CTRPsが運動によりどこに局在する脂肪組織から分泌増加しているのかは明らかでない。そこで本研究では、有酸素性トレーニングによって局在する脂肪から運動誘発的にCTRPs(adiponectin、CTRP3、CTRP5)が発現増加するか否かを検討することを目的とした。
本研究では、老化モデルマウスとしてSAMP1マウスを用いて、回転車輪を用いた自発走運動による有酸素性トレーニングを12週間実施し、12週間後に精巣上体、大動脈血管周囲、肩甲下皮下および褐色の脂肪を摘出した。精巣上体脂肪および肩甲下褐色脂肪のadiponectin 遺伝子発現は、安静対照群(Con群)と比較して運動群(Ex群)で有意に高値を示した。また、精巣上体脂肪のCTRP3遺伝子発現は、Con群と比較してEx群で有意に高値を示した。一方、CTRP5遺伝子発現は、すべての脂肪組織においてCon群とEx群との間に有意差は認められなかった。
これらの結果から、有酸素性トレーニングによるCTRPs遺伝子発現の増加は脂肪部位特異性がある可能性が示唆された。
