Outline of Annual Research Achievements

CRISPR/Cas9による座位特異的ノックインにより、標的遺伝子座位にレポーター遺伝子をノックインし、標的遺伝子の発現をモニターしつつ、ホモ接合体を作製し変異体の表現型を観察することが本来の本研究の目的であった。しかし、レポーター遺伝子のノックインはF0世代では成功したものの、F1世代を得ることは出来なかった。そのため、当該年度より、ノックイン系統作成の過程で得られたcart like遺伝子およびslurp-like遺伝子の変異体系統の表現型解析を行っていた。
これらの標的遺伝子とそのファミリー遺伝子をクローニングし、その発現をwhole mount in situ hybridization法により各発生段階において検出した。また、プラスチック切片の作製により発現部位を詳細に明らかにした。これらの遺伝子の進化系統樹を作成し、その保存性と機能ドメイン・発現部位について考察した。これらの変異体系統の表現型解析および標的遺伝子の発現解析の成果を、Spatiotemporal expression of the cocaine- and amphetamine-regulated transcript-like (cart-like) gene during zebrafish embryogenesis., Gene Expr Patterns.,2018およびDevelopmental expression of slurp-like1 and slurp-like2 during zebrafish early embryogenesis., Gene Expr Patterns.,2018として発表した。これらの標的遺伝子は未解析遺伝子であるため、発表論文が未解析の低分子量タンパク質の機能解析の進展の一助になることを願っている。
