Outline of Annual Research Achievements

自然界には、様々な物を食べる広食性の種と、特定の物のみを食べる狭食性の種が存在する。我々は、様々な栄養環境に対して個体がどのように応答し、適応(個体成長)を実現しているかを明らかにするため、食性の異なるショウジョウバエ近縁種に注目した。モデル生物キイロショウジョウバエは、自然界では発酵した多種類の果物を食べる広食性種で、適応できる栄養条件の幅が広い。一方で、その近縁種には特定の花や果実のみを食性とする狭食性種も存在する。そこで、キイロショウジョウバエを含む広食性二種と狭食性三種を選び、異なる栄養バランスに対する適応能力と応答を比較した。その結果、広食性二種はどの食餌条件においても蛹までの発生率(適応能力)が高い一方で、狭食性三種は低タンパク質かつ高炭水化物餌に適応できないことを見出した。そこで、これらの近縁種が野外で餌とする食物の成分を解析し、狭食性種の適応能力にとって鍵となる栄養バランス構成因子の絞り込みを行った。この結果をもとに、栄養バランスへの適応能力の種間差を生み出す全身性シグナル伝達経路を探索し、Activin シグナル経路に着目した。マルチオミクス解析の結果から、栄養バランスに応じた遺伝子群の発現調節が広食性種の代謝恒常性を維持し、その適応能力を支える可能性が考えられた。一方で狭食性種は、栄養バランスに対してキイロショウジョウバエの Activin 変異体と同様の遺伝子発現応答及び代謝産物量の変動を示した。この結果から、狭食性種は Activin シグナル経路内に変異が生じたために遺伝子発現制御ができず、低い適応能力を示すとの仮説を立て、その検証も行った。以上の全ての検証結果を論文としてまとめ、現在査読後の改訂版を再投稿する段階にある。
