Outline of Annual Research Achievements

本研究は肥満細胞の腫瘍化における細胞の恒常的増殖シグナルや、生体内における腫瘍の環境適応としての低酸素応答や代謝変化、免疫細胞との相互作用の解析を通して腫瘍細胞の生存戦略を明らかにすることを目指す。低酸素応答に関しては血管新生誘導因子（VEGF）産生について、その病態形成への影響を詳細に解析した。その結果、マウス炎症モデルだけではなくヒトの炎症性組織においても肥満細胞のVEGF産生が観察された。またVEGFは血管内皮細胞に対してだけでなく神経細胞にも直接作用する知見を得た。すなわち、肥満細胞は炎症性組織局所からの刺激を受けて血管誘導以外にも様々な細胞応答を引き起こし、病態形成に複雑に関与する可能性が考えられた。
上記解析により肥満細胞が末梢組織において低酸素状態にあることを示したが、一般に肥満細胞は脱顆粒反応によって多くの炎症性メディエーターを産生・放出する性質がある。肥満細胞が置かれるこの厳しい生体内環境、および脱顆粒に伴う大きなエネルギー喪失は細胞にとって強いストレスとなると想定されることから、各種ストレス応答が細胞に対して与える影響についてもあわせて解析した。その結果低栄養状態や低酸素により細胞増殖が抑制されること、また脱顆粒刺激により小胞体ストレス応答が誘導され、細胞の生存など複数の細胞反応を制御していることが明らかとなった。これは腫瘍化肥満細胞においても同様に細胞の生き残り戦略としてはたらいている可能性があり、腫瘍根治に向けた新たな戦略立案の一助となるものであると考えられた。
