| Outline of Final Research Achievements |
We found that BHLHE41 gene expression could suppress amplified RRM1 gene in anticancer drug resistant cell lines. We examined BHLHE41 about the function in tumor suppression and gene copy regulation. Doxycycline (DOX) inducing BHLHE41 could suppress lung cancer cell lines growth via autophagic cell death. Immunohistochemical study of clinical lung cancer samples indicate BHLHE41 protein expression is correlate with good prognosis of patients. We could established the cells those reproducibly RRM1 gene copy reducing by BHLHE41 expression.

| Academic Significance and Societal Importance of the Research Achievements |
がんで特有に起こる異常な遺伝子の増幅について解明し、がん遺伝子の増幅を抑制できればがんの治療につながる可能性がある。BHLHE41により再現よく増幅遺伝子のコピー数の減少を起こすことが確認でき、BHLHE41が遺伝子のコピー数を制御する機能を持つことが示せた。
また、BHLHE41の発現により肺がん細胞にオートファジー細胞死を誘導することを明らかにできた。肺がんの臨床検体での解析からBHLHE41の発現は肺腺癌の予後良好にかかわる分子であった。肺がんの発生早期にその発現が低下することが予想される結果を得ており、肺がんの発生機構の解明に道を開い。
