Outline of Final Research Achievements

In eukaryotes, RNA polymerase II requires general transcription factors to initiate mRNA transcription. We have determined the crystal structure of human TFIIE at atomic resolution. In addition, mutations on the residues involved in the interactions resulted in severe growth defects in yeast. These findings provide a structural basis for understanding the functional mechanisms of TFIIE in the context of PIC formation and transcription initiation.

Academic Significance and Societal Importance of the Research Achievements

本研究では、基本転写因子TFIIEの結晶構造を原子レベルの精度で明らかにした。このことにより、基本転写因子がどのようにRNAポリメラーゼを制御しているかについて、そのメカニズムの一端を明らかにすることができた。また、基本転写因子は、古細菌からヒトまで幅広く保存されたタンパク質群であり、その詳細な構造からその機能を明らかにすることで、生物に普遍的に存在するDNAからRNAへの転写機構について、学術的に貢献できたと考えている。
