Outline of Final Research Achievements

We addressed whether CXCR4, a chemokine receptor, acts as a lactoferrin receptor in human T-cells. In Jurkat-T cells, human immortalized T-cells, lactoferrin induced phosphorylation of STAT3 was antagonized by a CXCR4 inhibitor. However, lactoferrin stimulation does not mimicked many aspects of CXCR4 activation, including receptor dimerization, tyrosine phosphorylation. These observations suggest that role of CXCR4 as a lactoferrin receptor is cell-type specific.

Academic Significance and Societal Importance of the Research Achievements

ラクトフェリンは乳中の生体防御タンパク質であり、免疫賦活作用と過剰な免疫応答を抑制する機能を併せ持つ。そのメカニズムを解明する目的で、獲得免疫を制御する主要な細胞であるT細胞におけるラクトフェリン受容体の同定を行った。その結果、ケモカイン受容体の一種であるCXCR4が、株化T細胞において、部分的にラクトフェリン受容体として機能することを明らかにした。
