Outline of Final Research Achievements

We developed a next-generation sequencing (NGS)-based method for genome wide SNPs analysis of Helicobacter pylori strains (HPs) specific genomes using gastric wash sample (G-Scan method). Gastric cancer-specific candidate 71 HP gene SNPs was obtained from analysis using the remaining 4 cases of gastric cancer, 4 cases of gastritis.

Academic Significance and Societal Importance of the Research Achievements

胃洗浄廃液内に混在した複数種ピロリ菌の全ゲノム解析(G-Scan法)により、「胃がんに特異的なピロリ菌遺伝子異常(OMPs: outer membrane proteins)」を版権することができた。これによるピロリ菌構造変化が胃粘膜定着強固と持続炎症に関与し、癌化を促し得る」可能性が推測され、今後は「ヒトゲノム+ピロリゲノム異常を分子マーカーとし、「廃液」による超早期胃がん診断を実現す」ため、前向き臨床試験の実施および、ピロリ菌遺伝子異常(OMPs)による発がんメカニズム解析を行う予定である。これにより、廃液を用いるだけで胃がん診断が可能となる。
