Outline of Final Research Achievements

Inflammasome is a key component of the innate immune system in endocrine and metabolic disease. We identified E3 ubiquitin ligase ARIH2 in activated NLRP3 inflammasome. ARIH2 ubiquitinate NLRP3 and negatively regulates NLRP3 inflammasome activation in macrophages. Our findings suggest ubiquitin regulation system as a potential target for the treatment of endocrine and metabolic disease.

Academic Significance and Societal Importance of the Research Achievements

炎症機構の一つであるインフラマソームのユビキチン修飾を介した調節系の責任分子としてARIH2が働くことを明らかにした。ARIH2はインフラマソームの活性化機構に働くNLRP3というタンパクをユビキチン修飾することによって、インフラマソームの活性化を抑制する、つまり炎症反応の抑制を行なっている。本成果は、インフラマソームの関与する内分泌・代謝疾患などにおいてユビキチン修飾系が、治療標的となることを明らかにした。
