Outline of Final Research Achievements

We are aiming to explore unexpected molecular mechanisms underlying various diseases by using spontaneously-occurring mutant hamsters. In this study, we made use of BIO14.6 cardiomyopathic hamster which exhibits marked cardiac hypertrophy. By crossing normal and BIO14.6 hamsters, we created a new hamster line lacking βB1-crystallin protein.
This study will be a starting point to elucidate the functional roles ofβB1-crystallin in both normal and failing hearts.

Academic Significance and Societal Importance of the Research Achievements

これまでβB1-クリスタリンは、水晶体に特異的なタンパク質と考えられてきた。すなわち、水晶体のクリスタリンは分子量によりα、β、γの3種類の複合体に分類され、それぞれはいくつかの単量体から構成される。
しかし今後は、本研究で開発した新しい疾患モデル動物を活用することで、これまで想像すらされなかったβB1-クリスタリンによる心筋の機能制御という新しい基礎医学分野の開拓が期待される。その上、ヒトの死因の3分の1を占める心臓病に対する新しい遺伝子診断と治療戦略の開発を可能にする予想外の可能性も切り開かれることであろう。
