Outline of Annual Research Achievements

＜目的＞アフリカツメガエルの造血は主に肝臓で行われ、骨髄は脂肪で満たされているが、低温（5度）刺激により脂肪髄における造血が誘導される。本モデルは従来のモデル動物はで解析が困難であった「温度と造血、脂肪髄での造血」といった課題に対して有用だと考えられる。本研究では造血幹/前駆細胞を指標とし、低温時における造血関連分子の変化を明らかにする。
＜研究成果＞
1.造血の指標として未同定であったツメガエル造血幹/前駆細胞の検出系を確立した。作出したツメガエルMplモノクローナル抗体および血小板に相当する細胞である有核の栓球を認識するモノクローナル抗体（T12）を組み合わせ、フローサイトメトリー法により細胞を分画した。その結果、Mpl+T12-細胞の大きさを示す前方散乱光（FSC）の小さい画分を分取すると形態的に未熟な細胞が濃縮された。本細胞の多分化能および生着能をコロニーアッセイ、移植実験によりそれぞれ確認した。よって、Mpl+T12-FSClow細胞には高度に造血幹/前駆細胞が濃縮されることが明らかとなった。
2.低温刺激により骨髄中の造血幹/前駆細胞が増殖することを示した。Mpl+T12-FSClow細胞数は低温刺激により骨髄でのみ優位に増加した。さらに、造血幹細胞の増殖因子であるTPOの骨髄内での発現量が増加していることを確認した。
3.低温刺激により骨髄細胞が増殖することが示唆された。マイクロCTによる骨髄環境の変化を解析すると低温刺激により骨環境が変化していく所見を得た。低温暴露に伴う組織、および骨形成関連遺伝子の発現量の変化を解析したところ、低温刺激により骨細胞の分化能が増加することが示唆された。
