Outline of Final Research Achievements |
Uric acid has been reported to be neuroprotective and inhibits neuronal death in Parkinson’s disease model mice. In this study, we established double mutant mouse of novel Alzheimer’s disease model mice(AppNL-G-F/NL-G-F) and knockout mice of uricase, which metabolize uric acid to allantoin. We performed immunohistochemistry of amyloid β and Y-maze test using 7-8 months old mice. However, we did not observed inhibition of accumulation of amyloid βand inhibition of reduced cognitive function.

Academic Significance and Societal Importance of the Research Achievements |
痛風患者においてアルツハイマー病の発症率が低いという疫学的報告や、他の神経変性疾患モデル動物における解析から、尿酸や尿酸代謝関連物質あるいは代謝酵素がアルツハイマー病の病態に関わっている可能性は高い。本研究では尿酸値の上昇がアミロイドβの蓄積を抑制する可能性は低いことを見出したのに加え、尿酸値上昇モデルの妥当性や認知機能異常解析の適切なタイミング等、尿酸とアルツハイマー病の関連を解析する上での今後の課題が明らかとなった。適切な尿酸値コントロールのエビデンスを得るためには、さらなる検討が必要である。
