Outline of Annual Research Achievements

癌遺伝子中毒(oncogene addiction)の構成的な理解を目的として研究を進めた。遺伝子編集技術によりヒト正常乳腺上皮細胞MCF-10A細胞にKRas遺伝子、またはBRaf遺伝子に癌で見つかる活性化変異を片アレルに導入した細胞株を入手した。このKRas遺伝子、BRaf遺伝子をsiRNAやCRISPR/Cas9によりKD、またはKOしたが、細胞死は起きなかったことから、これらの細胞株においてはまだoncogene addictionが起きていないことが分かった。これらの細胞株はERKのリン酸化がやや上昇していたが、soft agarアッセイや増殖曲線、MEK阻害剤に対する感受性などを調べた結果、野生株と大差がないことが分かった。これらの結果から、oncogene addictionが起きるためには、癌遺伝子に変異が入るだけでなく、その後のなんらかの変化(エピジェネティックな変化など)が必要であることが示唆された。
一方、MCF-10A細胞に癌遺伝子変異を導入したKRas遺伝子を過剰発現させた細胞株を樹立したところ、これらの細胞株についてはsoft agarアッセイや増殖曲線などに対する応答が野生株よりも向上しており、一般的な癌に見られる特性が観察された。しかしながら、過剰発現させたKRas遺伝子をsiRNAによりKDしたとしても細胞死は観察されず、oncogene addictionは起きていないことが分かった。これらの結果から、癌遺伝子を過剰発現させると癌に特有の特性である細胞増殖や足場非依存性は獲得できるがoncogene addictionは起きないことが分かり、oncogene addictionが起きるためには、なんらかの変化が重要であることが示唆された。
