Research Abstract

(1)Helicobacterlori感染COX-2/mPGES-1トランスジェニックマスにおけるCOX-2阻割剤etodolacの胃発がん抑制効果
炎症や発がんにcyclooxygenase-2(COX-2)の過剰発現が深く関わっている。野生型(WT)マウス,及びCOX-2とmicrosomal prostaglandin E2 synthase-1(mPGES-1)を過剰発現するトランスジェニック(Tg)マウスにHelicobacter pylori(Hp)感染,N-methyl-N-nitrosourea(MNU)投与,Hp+MNU投与の処置を行い,またetodolacによる胃発がんの抑制を検討した。52週で屠殺した結果,幽門腺部において,MNU群(Tg,15/15;WT,12/12),Hp+MNU群(Tg,19/19;WT,24/24)で全例で胃がんの発生が認めた。胃底腺部では,Tgマウス(MNU投与群:4/15,26.7%;Hp+MNU群:4/19,21.1%)でのみ胃がんの発生を認めた。etodolac投与群では,幽門線部胃がんの発生が有意に減少した(Tg,P<0.01;WT,P<0.05)。Tgマウスは,近年,欧米で増加している近位部胃がんのよい動物モデルと考えられた。また,etodolacはマウス胃発がんに対して抑制効果を有することが示された。
(2)骨髄由来幹細胞のHelicobacter pylori感染マウス胃発がんへの関与
慢性胃炎において誘導される骨髄由来幹細胞が胃腫瘍の起源になりうると報告されたが,がん化に如何に関わるか明らかではない。そこで,雄Green mouseの骨髄をX線を照射した雌C57BL/6Jマウスに移植し,HpとMNUを投与した。今後,発生したがんの細胞系譜を検討する予定である。
