Research Abstract

上皮間葉転換は運動能、浸潤能の亢進を伴うため、がんの浸潤能獲得に深く関わっている。また、上皮間葉転換はE-カドヘリンを介した細胞間接着をもつ細胞から、強固な細胞-細胞外基質接着を持つ細胞への変換と見ることができる。細胞接着による細胞挙動の調節にはインテグリン裏打ちタンパク質やカドヘリンの裏打ちタンパク質が重要であることがわかっているが、上皮間葉転換におけるこれらの機能は詳細には解析されていない。これまでの我々の研究からインテグリン裏打ちタンパク質ビネキシンの結合タンパク質が上皮間葉転換を抑制することが分かってきた。そこで、今年度は、本タンパク質による上皮間葉転換の制御メカニズムを解明し、新しい抗がん剤開発のための基盤的知見を得ることを目的とした。まず、TGFβの刺激により上皮間葉転換を誘導すると、本タンパク質の発現が減少することが確認できた。また、TGFβの刺激により活性化するシグナルを同定し、そのシグナル伝達の阻害剤や阻害タンパク質の上皮間葉転換への効果を調べた。このタンパク質の発現抑制による上皮間葉転換は、一部の阻害剤と阻害タンパク質により抑制された。一方で、このタンパク質の発現抑制が、E-カドヘリンの細胞表面への分配に影響するという証拠は得られていない。これらの結果から本タンパク質がTGFβ経路の一部のシグナルを調節することで上皮間葉転換を抑制していることが示唆された。
