Research Abstract

重粒子線照射による癌増殖抑制機構について解析し、患者個別のがん情報を得る方法を開発する事を目的として研究を行った。このため重粒子線治療で著しい局所制御の観られる悪性黒色種培養細胞を用い、重粒子線特異的に応答する遺伝子を検索した。重粒子線で発現抑制された遺伝子中、X線に比べ応答が有意な遺伝子には細胞周期関連遺伝子(CCNA2,CDCA8,WEE1など)が多く含まれており,炭素線の生物効果には,細胞周期の調節機構が重要な役割を担っている可能性が示唆された。染色体切断の再結合は重粒子線ではX線などに比べて低効率であり、再結合後の残存染色体異常数と細胞致死の効率は良い相関を示すものの、残存異常数に対する突然変異の効率は重粒子線では高かった事から、修復の質が異なり重粒子線では誤修復の割合が多いかも知れないことが示唆された。核酸誘導体トレーサーの集積動態は血流補正を行うことにより、インビトロで得られた低酸素による放射線抵抗性発現と同程度に検出できることを明らかにした。ヒトグリオーマ移植マウスでは^<18>FDGが最も高い集積を示し、投与直後の分布は血流分布およびDNA合成亢進部位と一致していたが、後期画像では低酸素部位への高度の集積をみとめた。また14Cベンジルアセテートの腫瘍内分布は他のプローブと比較して均一な分布をしめした。^<61>Cu,^<62>Cu標識ATSMについては、実用的な製造法を確立して安全性試験を行い、臨床利用のための承認を受けて臨床研究が開始された
