Research Abstract

我々はこれまでにフィーダー細胞を用いずに、ES細胞の細胞塊を無血清下に浮遊培養ですることで、効率よく神経細胞に分化させる系をまず樹立した(SFEB法)。マーカー解析の結果、SFEB法でES細胞から産生された神経細胞はこれまで産生が困難であった大脳の前駆細胞であることが明らかになり、さらにShhを作用させることにより、この大脳前駆細胞から大脳基底核などの細胞を試験管内で分化誘導することに成功した。この研究により、従来不可能であった試験管内での大脳神経細胞の大量産生が可能なり、大脳の変性疾患(ハンチントン病やアルツハイマー病など)の発症機序の解明や大脳疾患の治療法開発に大きく貢献することが期待される。SFEB法によりES細胞からの前脳の分化誘導は確認されたが、小脳、橋などへの分化効率は低い。小脳、橋などを含む変性疾患に関連する後脳吻側部に着目し、細胞外シグナルによる分化誘導系の樹立を目指し、条件検討を行い、マウスES細胞からの10%程度での小脳主要ニューロンの安定した分化誘導法を確立した。一方、ヒトES細胞分化にSFEB法を応用するにあたっては、ヒトES細胞特有の問題として、細胞死による細胞生存の低さがあった。今年度、この細胞死をROCK阻害剤が抑制することを発見し、大量培養を効率よく行うことが可能となり、これを用いてヒトES細胞からの大脳の分化誘導にも成功した。ヒトES細胞の技術をさらに広く再生医療や創薬に用いるために重要な基盤技術が確立された。
