Research Abstract

長期血液透析患者の骨関節組織に沈着するβ2-ミクログロブリン(β2-m)アミロイド線維は、従来試験管内ではpH2.5という酸1生域でしか伸長せず、中性pHでは脱重合していた。最近、特定の生体分子群がβ2-mアミロイド線維(fAβ2M)に特異的に結合し線維を安定化させること、臨界ミセル濃度程度のドデシル硫酸ナトリウム(SDS)が中性pH域でβ2-mの立体構造を部分的に変化させ、線維伸長を引き起こす事を解明した。本研究では、SDSと同様の効果を有する生体分子を探索し、生体内線維形成機序を解明する事、及び線維形成を阻害・不安定化させる有機化合物を探索し、治療薬開発への糸口をつかむ事を目指す。これにより、病態解明の進展と、新たな治療戦略の構築が期待できる。本年度の成果は下記の通りである。
1.SDSに類似した構造を持つ生体分子である各種リゾリン脂質のfAβ2M伸長効果を、脂肪酸炭素数と親水基について系統的に解析した結果、マイナスに荷電した親水基を持っリゾフォスファチジン酸とリゾフォスファチジルグリセロールは、fAβ2Mの伸長促進効果を示すことを確認した。また、ラウリン酸、オレイン酸など一部の遊離脂肪酸にも線維伸長促進効果があることを見出した。
2.透析患者から抽出したアミロイド線維を核として、線維がSDSの添加により中性pH溶液中で伸長することを示し、生体のアミロイド中にSDS型のシードが含まれていることを確認した。また、SDS型と酸性pH環境下で伸長する線維の立体構造が異なっている可能性があることを赤外吸収スペクトル法などにより示した。
3.ポリフェノール類などの有機化合物を試験管内線維伸長反応系に添加し、線維伸長の阻害能を探索した。チオフラビンT蛍光法、電気泳動法などにより、阻害活性を示す物質を見出した。
