Research Abstract

有機結晶は構成する分子の立体構造が規制された特異な反応場を提供するため,不斉反応,立体規則的な反応などの合成化学的な立場から幅広く研究されている.ところが,固体-固体,固体-気体,固体-液体などの不均一系反応に応用した場合や反応に大きな原子移動を伴う場合には,全く反応が進行しないか,または反応の進行に伴い結晶構造が崩れてしまう.結晶相反応は反応の選択性に優れているが,一方では反応への制限が多いことを示唆している。このジレンマを克服するために,本研究では結晶を構成する分子の挙動をエントロピー制御することで,結晶化により記録された分子情報を記憶して,このキラルメモリーを様々な高選択的反応へ展開することを目的とし研究した.
(成果1)アキラルなクマリンカルボン酸アミドの不斉結晶化により発現した不斉を光増感反応へと展開して99%eeの光学純度で光不斉反応を達成した。同様にして、硫黄イリドとの反応では不斉シクロプロパン化をカルベンとの反応ではジヒドロフラン環の不斉合成に成功した。これまでに例のない新しい手法による絶対不斉合成である。
(成果2)上記の手法をヂアステレオ選択的光不斉反応へと応用した。複数のジアステレオマー混合物として存在する分子のエピ化優先晶出法により単一のジアステレオマーに完全分割し,エントロピー制御された環境での高選択的不斉反応を達成した。アミド基に不斉源としてプロリンを有するクマリンカルボン酸アミド、2-アルコキシ1-ナフトアミドと2-キノリノン-4-カルボン酸アミドを一度結晶化させてから低温で溶解し反応させることによりいずれの場合もほぼ100%deの光不斉反応に成功した。
