不斉・交差チタン=クライゼン・アルドール反応を利用する特異機能分子の実用合成

Research Abstract

従来塩基法より高い反応性,特異な選択性を有するTi-Claisen縮合の研究を行っている.これまで事実上困難であった酸クロリドとメチルエステル間の交差型Ti-Claisen縮合^<1)>さらにケテンシリルアセタールとメチルエステル間の塩基触媒交差型Claisen縮合を最近報告した^<2)>.
最近,初めての不斉Ti-Claisen縮合を開発した.すなわち,入手容易な光学活性アトロラクチン酸誘導体であるcis-ジオキソラン-4-オンを不斉補助基として用いる方法^<3)>を見出した.
この化合物のテンプレート効果を活かし非常に高いジアステレオ選択性(〜99%de)でアシル化体を得た後,引き続くメタノリシスで目的とする光学活性α-ヒドロキシ-β-ケトエステルを合成することに成功した.不斉補助基は入手容易であり,しかも反応後ラセミ化することなく,簡便な方法でほぼ完全に回収可能である.
この反応を活かし,植物毒素Alternaric acidの短段階形式不斉全合成を達成した.また現在,血管新生抑制剤として注目されている(-)-Azaspireneの不斉全合成を進行中で,今後,独自の不斉交差Claisen縮合・aldol・Mannich反応の開発に注力し他の複雑な構造を有す鎖状生理活性天然物合成,さらには構造活性相関解明への展開も計る.
