Research Abstract

BTEB2はzinc finger型の転写因子であり、胚性幹細胞(ES細胞)、栄養胚葉幹細胞(TS細胞)、始原生殖細胞、また成体においては、腸上皮、皮膚上皮において豊富に発現し、幹細胞や生殖系列にかなり限局して発現している。BTEB2遺伝子欠損ES細胞は樹立可能であり、未分化性を維持しているが、細胞増殖速度に著名な減少を示す。逆にBTEB2を強制発現させたES細胞は、LIFを除去して分化誘導を行っても分化せず、未分化性を維持し得ることを明らかにしている。しかしながら、BTEB2が幹細胞の増殖および未分化性をどのように制御するのか分子レベルでは不明である。
BTEB2による幹細胞の増殖制御機構を分子レベルで理解するためBTEB2野生型ES細胞、BTEB2欠損ES細胞、BTEB2過剰発現ES細胞からmRNAを単離し、マイクロアレイ解析によって標的候補遺伝子を推定することを試みた。マウスジーンチップを用いて43,000転写産物の発現変化を検討したところ、BTEB2野生型ES細胞に比べて、BTEB2欠損ES細胞において発現が減少し、BTEB2過剰発現ES細胞においては上昇している転写産物群、逆にBTEB2野生型ES細胞に比べて、BTEB2欠損ES細胞において発現が上昇し、BTEB2過剰発現ES細胞においては下降している転写産物群を得た。その中で、ES細胞の増殖に関与していることが報告されている遺伝子Tcl1についてReal-time PCR法により遺伝子発現の変化を検討したところ、マイクロアレイの結果と一致する結果を得た。更に、Tcl1の標的であるAkt1(PKB)のリン酸化状態を検討した所、顕著な変化を受けていることを明らかにした。
