B細胞分化における免疫グロブリンの品質管理とその破綻

Research Abstract

昨年度の研究で確立した、in vitroでのプレB細胞レセプター(preBCR)発現による骨髄プロB細胞の分化誘導システムを用いて、「品質」の異なる種々のIgHμ鎖をμ鎖欠損マウス由来の骨髄プロB細胞に発現させた。導入したμ鎖の代替L鎖との結合能に比例して、preBCRの発現レベル、preBCRからのシグナル強度、プレB細胞の増殖・分化の程度に違いが認められたが、preBCR発現の経時変化は酷似していた。すなわち、preBCR発現は一過性で、μ鎖遺伝子導入後1日目をピークとして、いずれの場合も3日目には発現が消失した。これまでの研究では、preBCRからのシグナルによって代替L鎖遺伝子の転写が抑制される結果preBCRの発現が終焉するとされてきたが、本研究での解析から、転写抑制がおこる以前にすでに蛋白レベルでpreBCRの発現抑制がおきていることが判明した。preBCRのシグナル伝達にかかわるBLNKを欠損するマウスでは、プレB細胞上でのpreBCRの発現停止がおこらず、プレB細胞の白血病化を誘導することが示されている。したがって、プレB細胞の増殖制御には、蛋白レベルでの素早いpreBCRの発現制御が必要なものと考えられる。そこでBLNK欠損プレB細胞株に、発現誘導型のBLNK発現ベクターを導入して、preBCRの発現終焉を誘導できる系を樹立した。その結果、preBCRからのシグナルにより、代替L鎖の転写抑制以前に、preBGRの細胞内への取り込み(internalization)が促進し、細胞表面でのpreBCR発現終焉がこおることがわかった。このpreBCRの発現制御の破綻によりプレB細胞が白血病化すると考えられることから、この系を用いて、現在その分子機構を解析している。
