Research Abstract

本年度は固体高分解能NMR分光法の手法を用いて化学シフト相互作用や磁気相極子相互作用などの異方的磁気相極子相互作用を利用して膜結合生体分子の立体構造や配向を精密に決定するための方法論の開発を行った。さらにこの方法を用いて膜タンパク質など膜結合生体分子の構造と機能の相関を分子レベルで明らかにする研究を行い以下の成果を得た。
バクテリオロドプシン(bR)はHalobacterium salinarumの紫膜に存在する内在性膜タンパク質でありシッフ塩基結合によってレチナールはLys-216と共有結合している。bRは7本の膜貫通ヘリックスから構成されており、光駆動型プロトンポンプ活性をもっている。高速MAS条件下での圧力および光摂動によるバクテリオロドプシン(bR)におけるレチナールの異性化とタンパク質の構造変化の解析を行った。本研究では[ζ-^<15>N]Lys-bRの^<15>N CP-MAS-NMRスペクトルを観測して、圧力によるレチナールの異性化を観測した。この結果、暗順応状態ではall-transと13-cisが等量で存在し、圧力順応状態では13-cis状態が増加し、明順応状態ではall-trans状態が増加することが判明した。
Pharaonis phobirodopsin(ppR)はtransducer(pHtrII)と複合体を形成しており、負の光走行性を示す信号伝達機能をもっている。このタンパク質複合体のC-末端およびループ部位についてDD-MAS-NMR法で観測し、膜貫通部位はCP-MAS-NMR法により観測した。この結果、暗順応状態では細胞質側のpHtrIIはppRのC-末端と相互作用をもっているが、光活性化状態であるD75Nとの複合体ではC-末端とpHtrIIとの相互作用は弱くなることが判明した。さらにpHtrIIの信号を観測したところ、光活性状態ではpHtrII同士で相互作用が起こり、相互作用のスイッチングの起こることが判明した。
