Research Abstract

ゲノムはDNA二重鎖切断による死と修復による再生をたえず繰り返している。この選択にかかわっている大腸菌のRecBCD酵素は、DNAの切断点からDNAに取り付き、ATPを大量に消費してDNAを巻き戻し、分解しながら進んでいく。このとき、RecBCD酵素はカイ配列というゲノムのID配列に出会うと、分解を停止して基質DNAを相同組換えで中心的な役割を担うRecAタンパクに引継ぎ、DNAの修復を開始させる。このゲノム維持ナノマシーンの反応機構の詳細を、「ゲノムのID配列に出会った時に何が起きるか」に焦点を絞って明らかにしてきた。まず、生化学的手法と一分子可視化手法を組み合わせて研究を行う環境づくりを行い、我々の研究室において変異タンパクの発現と精製が効率よくできる実験環境を整備した。
ゲノムのID配列とそれを認識し分解酵素から修復酵素へと切り替わる仕組みは、細菌の種を越えて保存されている。大腸菌ではRecBCD酵素はRecB, RecC, RecDという3種のサブユニットから構成されているが、遠縁の枯草菌では2つのサブユニットから構成される類似酵素が同様の反応を行う。大腸菌のRecBCD酵素を用いた-分子可視化実験によって明らかにしたゲノムID配列上での酵素の「停滞」という現象を、この枯草菌アナログと枯草菌ゲノムID配列でも生化学的に示すことができた(J.Biol.Chem.2006)。このことは、この酵素の制御配列上での停滞がサブユニット組成によらず、この酵素-DNA相互作用において普遍の現象であることを示唆していた。また、このゲノムのID配列を認識できない変異RecBCD酵素において、それが認識する新規ID配列での相同組換えを促進するRecA変異を発見し、生化学実験と遺伝学実験によってこれを解析した(J.Mol.Biol.2007)。
