Research Abstract

様々な環境条件での遺伝子発現を、モデル生物のラン藻Synechocystis PCC6803のDNAマイクロアレイを用いて解析し、光合成生物にのみ広く保存され且つ植物では葉緑体に局在すると予想された、約100個の機能未同定遺伝子を抽出した。これらの機能未知遺伝子は、特定の環境ストレス条件下で葉緑体機能の分化・環境適応に、極めて重要な役割を果たしていると考えられる。本研究では、これらの未知遺伝子にコードされたタンパク質の機能を解析し、葉緑体の環境ストレス適応過程における役割を明らかにするとともに、植物個体レベルでの生存戦略における役割を明らかにすることを目的とする。
本年度までに、Synechocystisで作製したストレス応答性機能未知遺伝子の変異株約90種について、通常の培養条件と環境ストレス条件で光合成活性を測定した。その結果、slr1674遺伝子の変異株で、酸化ストレスなどの環境条件下で、顕著な光合成活性の低下が観察された。Synechocystisにはslr1674のパラログ遺伝子slr1638も存在する。slr1674はストレス誘導性で、slr1638は構成的に発現する遺伝子であった。しかしながら、どちらの変異株もストレス条件下で野生株に比べ生育速度が低下する表現型を示した。いくつかのラン藻ではこのホモログ遺伝子がリーダーペプチダーゼ遺伝子lepBと共転写されること、Synechocystisの発現プロファイリング解析の結果slr1638の発現がlepBと類似していることが分かり、膜移行・膜局在タンパク質の成熟化に関わる可能性が示唆された。そこで、チラコイド膜ルーメンに移行するPsbUタンパク質の蓄積量を比較したところ、両変異株では野生株に比べてPsbUの蓄積量が少ないことがわかった。
