Research Abstract |
In this research, a new technology for creation of nano-mechanisms using DNA and related biotechnology has been investigated and obtained the following results. 1.Periodic construction of nano-mechanisms by DNA: Nano-particle modified DNA double strand fractions have been created and ligated (connected each other by enzyme) for several tens of blocks. Since the pitch of the DNA nucleic acid is 0.34nm, the positioning resolution is very fine compare to those of mechanically obtained. Experimental results show the periodic nano-structure by the optimization of the sequence of the nucleic acid, which are observed by the AFM. 2.Creation of Molecular nano-mechanisms by protein complex: To add the function such as actuators and sensors, a complex with DNA and protein gas been proposed. A DNA folding protein called IHF has been introduced to bend the DNA at designated nucleic sequence, and experimentally confirmed by AM micrographs. Also, a protein ReccA has been introduced to branch DNA at the hybridized sequence, which is experimentally confirmed as nano-junctions. 3.Nano-structures by circular DNA and Mechanical Property Evaluation of DNA : Three types of nano-structure, DNA glasses, Serial DNA rings, and DNA chain has been proposed and experimentally fabricated. A DNA glasses with a diameter of 27nm has been formed and confirmed by AFM. A mechanical model of the DNA has been proposed by combining coil spring for main structure and small springs for nucleic and hydrogen bonding, which is compared with the experimental results and show good agreement.
