Research Abstract

A:染色体外遺伝因子の、微小核への取り込みを介した細胞外排出HeLa H2B-GFP細胞を用いて集中的なタイムラプス実験を繰り返すことにより、染色体型微小核の形成機構、微小核を持つ細胞の挙動、および、微小核多様性の原因について新発見が多く得られた。また、我々はすでにDMを生細胞内で可視化したCOLO 320DM-GFP株を樹立していたが、本年度にはこの細胞を用いて周到なタイムラプス実験を行うことにより、間期核からDMが細胞質に移動して微小核を形成する過程、および微小核が細胞質膜のブレッブに取り込まれて細胞外に放出される過程、について明確な根拠を得ることができた。このことは細胞生物学上極めて重要な発見であるとともに、DMが細胞間で転移する可能性を示唆しており、重大な意味を持つ。(B;染色体外遺伝因子を介した遺伝子増幅の機構)HSRやDM内での組み替え点を極めて多数単離し、その塩基配列を決定していくことにより遺伝子増幅の分子機構に関する理解が深まった。一方、末端がヘアピン構造である直鎖状プラスミドDNAは、IRとMARの配列を持つことに依存して、染色体外で安定に維持されるようになることを前年度に見いだしていた。本年度には、その分子機構に関して明確なモデルを得ることができた。この実験系は、染色体外で複製開始を指示する配列を評価することができるため、極めて有益な系である。(C;遺伝子増幅を利用した蛋白質生産)IR/MARプラスミドとテロメア配列を組み合わせることにより、染色体外で増幅して安定に維持され、なおかつ蛋白質発現量が高くなる新規実験系を見いだした。この方法をテロメア化IR/MAR遺伝子増幅法と命名した。この方法は、細胞生物学上興味深いとともに、蛋白質生産にとっても有力な方法となることが期待された。
