Research Abstract

Current study was undertaken to develop a low invasive and high efficient angiogenic cell and gene therapy for the ischemic heart diseases by a transplantation of de-differentiated fat (DFAT) cells transfected with VEGF gene. We evaluated a differentiaon of DFAT cells to the vessel and cardiomyocyte in culture. DFAT cells affiliated the vessel formation of HUVEC, and have abilities of differentiation to endothelial cells and vascular smooth muscle cells. We created the myocardial infarction in adult pigs and transfected VEGF gene into DFAT cells by HVJ-envelope. We transplanted DFAT cells with VEGF into the infracted hearts using a double catheter planted in coronary vein retrogradely. The transplantation of DFAT cells with VEGF gene significantly improved cardiac functions and increased vessel number in the infracted hearts compared to transplantation of DFAT cells without VEGF gene. For the clinical application, we evaluated an efficient and low invasive methods to prepare DFAT cells. Moreover, we established strategies for the standard culture and the banking of DFAT cells in GMP levels.
