Research Abstract |
For producing the ploidy periclinal chimera in Citrus species, in vitro grafting using the microsurgery method was examined. Hypocotyl segments of in vitro seedlings of diploid and tetraploid Meiwa kumquat (Fortunella rassifolia Swingle) were used. After a month of grafting, a lot of shoots were regenerated from upper region, and the frequency was the highest (95%) in 1/2 Murashige and Skoog medium containing 1 mg/16-benzylaminopurine. However, the ploidy of the regenerated shoots was diploid or tetraploid, and there were no ploidy periclinal chimeras. Whereas, in vitro grafting between GUS gene-transformed diploid plant and tetraploid plant was carried out, for investigating the origin of ploidy periclinal chimeras. Although transgenic plant-derived callus and shoots were detected by the staining, chimeric tissue could not obtain.
