Research Abstract |
1) Hamster intracytoplasmic sperm injection (ICSI): (1) Hamster oocytes injected with hamster sperm heads developed into morulae, but did not develop to blastocysts in albumin-free HECM-9 medium. Supplement of human serum albumin (0.5 mg/m1) to HECM-9 medium promoted blastocyst development (36%). (2) Supplement of recombinant human albumin to HECM-9 medium promoted blastocyst development (26%) as well with HSA. (3) Supplement of hemoglobin (1-10 μg/ml) to HECM-9 medium did not improve blastocyst development. (4) Although addition of 0.5 mM glucose to HECM-9 medium did not affect embryonic development, implantation rates of embryos developed in glucose-containing medium was higher than those in glucose-free medium. However, fetal development was the same. 2) Hamster round spermatid injection (ROSI) : (1) Although hamster oocytes injected with round spermatids (ROSI) were normally fertilized (54% vs. 60%) and developed to blastocysts (14% vs. 13%) at the same rates regardless of activation treatment, fetal rate of activated ROSI-embryos (22%) was significantly higher than that of non-activated ROSI-embryos (3%). (2) Hamster oocytes injected with fresh and frozen-thawed round spermatids developed to blastocysts at the same rate. 3) Cat ICSI and ROSI : (1) In vitro -maturation of cat oocytes was depended on the grade of cumulus-oocyte complex. Maturation rate in the A grade was higher than that in the other grades. (2) In vitro matured-cat oocytes injected with cat spermatozoa developed to blastocysts without additional activation (25%). (3) In vitro-matured cat oocytes injected with round spermatozoa did not developed to blastocysts without additional activation, but two times of 5 μM ionomycine for 5 min and followed by 3-h culture in 1.9 mM 6-dimethylaminopurine stimulated blastocyst development (28%).
